Mutations in somatic cells are a hallmark of cancer. The resulting mutant proteins are processed into mutant peptides, which are then presented to the surface of tumor cells by major histocompatibility complex (MHC) I molecules and captured by T cells. When recognized, an anti-tumor immune response is triggered, and this mutated epitope peptide is called a neoantigen. Using modern gene deep sequencing methods, a large number of somatic mutations in tumors can be identified, combined with modern proteomics and bioinformatics techniques, to screen out tumor neoantigen polypeptide gene sequences that may be recognized by T cells and have strong immunogenicity.
